An extensive introduction of alanine residues was carried out by Zhang who placed up to five alanines in the 126-134 alpha helix of T4 lysozyme. The substitution of alanine at the Glu128,Val131 and Asn132 increased the thermostability of T4 lysozyme. The increse in stability at the three sites was found to be additive.
