The enzymatic reaction of serine protease has two major phases:

• Acylation in which the peptide bond is cleaved and an ester linkage is formed between the peptide carbonyl carbon and the enzyme.
• Deacylation in which the ester linkage is hydrolized and the enzyme regenerated.
